Study of antigen Test from Blood group 

Principle:   

  1. Rh+ and Rh- agglutinogen in RB.C. man are called as Rhesus factor. 
  2. Name Rhesus factor has been derived from short-sized Marnosar Rhesus macaque monkeys.  
  3. If red cells of Rhesus monkey are injected into the blood of rabbit. Then it is observed that 86% R.B.c. of rabbit agglutinate showed clumping and 14% R.B.C. remain unaffected. The same reaction is shown by the blood of Rhesus monkey and man. 
  4. The agglutinating blood is called as Rh+ while non-agglutinating blood is called as Rh-.Because of presence of D-antigen on R.B.C. of man, blood group may be Rh+.
  5. In the absence of D-antigen blood group is Rh-. In persons RB.C. having D-antigen is agglutinated by antibodies.
  6. Such blood is Rh+. In absence of D-antigen in RB.C., blood is Rh-. or Rh+ blood groups
  7. Take a clean glass slide. 
  8. Add a drop of Anti-D serum on the slide and then add a drop of blood left for 2 minutes and then observe observation and result. If agglutination occurs it

 Slide examination method

Procedure: 

  1. Take a clean glass cavity slide. 
  2. Add a drop of Anti-D serum on slide 
  3. Then add a drop of blood leave for 2 minutes and then observe observation and result. 
  4. If agglutination occurs it indicates that D-antigen in serum has resulted in clumping and blood in Rh +.
  5. If no clumping blood in Rh-.
